A simple, accurate, sensitive and reliable direct Spectrophotometric method have been developed, for the determination of Lansoprazole (LPZ) either in pure form or in pharmaceutical formulations by complex formation with Bromocresol Green (BCG). The method is based on the interaction of LPZ in dichloromethane (DCM) medium with acidic sulfonephthalein dye, namely, bromocresol green (BCG) to form stable, yellowcolored, ion-pair complex peaking at 417 nm. The variables were studied in order to optimize the reaction conditions. Beer's law was obeyed in the concentration range of 0.250-20.00 μg.mL-1 with good correlation coefficient (R2= 0.9999). The relative standard deviation did not exceed 3.97%. The limit of detection (LOD) and the limit of quantification (LOQ) were 0.033 and 0.101 μg.mL-1, respectively. The molar absorptivity was calculated to be 2.3068×104 L.mol-1.cm-1. The composition of the ion-pair complex was found to be 1:1 by Job's method of continuous variations and the conditional stability constant (β) of the complex has been calculated. The proposed method was successfully applied to the analysis of Lansoprazole either in pure form or in pharmaceutical formulations with good accuracy and precision. The results obtained by the proposed method were compared favorably with those of the reference method.Keywords

A simple, direct and accurate spectrophotometric method has been developed for the determination of cefpodoxime proxetil (CEFP) in pure and tablet dosage form. The method involves the formation of yellow ion-pair complexes between the cited drug and bromophenol blue (BPB) using some solvents. Dichloroethane and chloroform were the best. The absorbance is measured at λmax 412 and 422 nm in dichloroethane and chloroform, respectively. Variables were studied in order to optimize the reaction conditions. Molar absorptivity (ε) for [CEFP]:[BPB] and [CEFP]:[BPB]2 complexes were selected. Beer's law was obeyed in the concentration range of 0.2788-44.608 μg.mL-1 and 0.5576-44.608 μg.mL-1 in presence of 1x10-3 mol.L-1 of BPB with good correlation coefficient (R2=0.9996 and R2=0.9998) in dichloroethane and chloroform, respectively. The relative standard deviation did not exceed 4.3%. The proposed method was validated for specificity, linearity, precision and accuracy, repeatability, sensitivity (LOD and LOQ) and robustness. The developed method is applicable for the determination of CEFP in pure and different dosage forms with average recovery 98.8% to 101.1%, and 98.6% to 101.2% in dichloroethane and chloroform, respectively, and the results are in good agreement with those obtained by the RP-HPLC reference method.Keywords

A simple, direct and accurate spectrophotometric method has been developed for the determination of cefpodoxime proxetil (CEFP) in pure form and pharmaceutical formulations by complex formation with bromocresol purple (BCP). The method involves the formation of yellow ion-pair complexes between BCP reagent and CEFP in chloroform. The two formed complexes ([CEFP]: [BCP] and [CEFP]: [BCP]2) have maximum absorption at λmax 414 nm. The proposed method was validated for specificity, linearity, precision and accuracy, repeatability, sensitivity (LOD and LOQ), robustness and solution stability with an average recovery of 99.2-101.0%.The formed complexes ([CEFP]:[BCP] and [CEFP]:[BCP]2) were measured against the reagent blank prepared in the same manner. Variables were studied in order to optimize the reaction conditions. Molar absorptivity (ε) for two complexes were 10500 and 15700 L.mol-1.cm-1, respectively. Beer's law was obeyed in the concentration range of 0.5576-5.576 and 5.576-55.760 μg/ml in the present of 2x10-4 and 1x10-3 mol/l of BCP, respectively, with good correlation coefficient (R2= 0.9988 and R2= 0.9995, respectively). The relative standard deviation did not exceed 4.4%. The limit of detection (LOD) and the limit of quantification (LOQ) were 0.083 and 0.25 μg/ml, respectively. The developed method is applicable for the determination of CEFP in pure and different dosage forms with the average assay of marketed formulations 91.6 to 106.2%, and the results are in good agreement with those obtained by the RP-HPLC reference method.Keywords

Two simple, direct and accurate spectrophotometric methods have been developed for the determination of Glimepiride (GLM) in pure and pharmaceutical formulations by ion-pair complexes with Bromophenol Blue (BPB) and Thymol Blue (TB). The methods involve the formation of yellow ion-pair complexes between BPB and TB with GLM at pH<3.8; after reacting GLM with Na2CO3 to give C24H33N4H+O5NaS which is prepared in chloroform. The formed complexes [GLM]:[BPB] in chloroform and [GLM]:[TB]2 in acetonitrile were measured at λmax 424 and 392 nm, respectively. Molar absorptivity(ε) for complexes were 23000 and 28000 L.mol-1.cm-1, respectively. Beer’s law was obeyed in the concentration range of 1.126 – 46.608 μg.mL-1 with relative standard deviations (RSD) ≤ 3.8% in presence of 5.0x10-4 mol.L-1 of BPB and 0.319 – 34.343 μg.mL-1 with RSD ≤ 4.0% in presence of 2.5x10-4 mol.L-1 of TB. The proposed method was validated for specificity, linearity, precision, accuracy, repeatability, sensitivity (LOD and LOQ) and robustness.
The results for the determination of GLM using TB were more sensitive and accurate than using BPB. The developed method is applicable for the determination of GLM in pure and different dosage forms with average assay of 99.0 to 101.0% and the results are in good agreement with those obtained by the RP-HPLC reference method.
